Simultaneous Detection of Foodborne Pathogens Using a Real-Time PCR Triplex High-Resolution Melt Assay

چکیده
Foodborne pathogens pose risks to populations all over the world. Pathogens can be used as bioterrorism agents, causing an outbreak that affects many individuals through consumption of a commonly affected food or beverage. A PCR assay identify their unique melting points using high-resolution melt assay. Assays detect bacteria individually from mixture species-specific primers. An was developed and three routinely cause multistate foodborne outbreaks, documented by U.S. Centers for Disease Control Prevention, Campylobacter jejuni (C. jejuni), Escherichia coli (E. coli), Salmonella enterica (S. enterica), in single bacterium assays multiplex. The primers were targeted specific gene sequences each pathogen, including cadF, yedN, hilA, respectively. Each pathogen identified its temperature assays: 78.10 ± 0.58 °C C. jejuni, 81.96 0.42 E. coli, 87.55 0.37 S. enterica. multiplex successfully detected with distinctly separated peaks. also proved specific, reproducible, fast, sensitive experiments.

Detection and discrimination of two Brucella species by multiplex real-time PCR and high-resolution melt analysis curve from human blood and comparison of results using RFLP
Objective(s): Rapid and accurate detection of Brucella abortus and Brucella melitensis from clinical samples is so important because antibiotic treatment has major side effects. This study reveals a new method in detection of clinical samples of brucellosis using real-time PCR and high-resolution melt (HRM) curve analysis. Materials and Methods: 160 brucellosis suspicious samples with more tha...

doi: 10.1111/j.1757-837X.2011.00113.x Abstract Introduction Salmonella, Shigella and Escherichia coli O157: H7 are major foodborne pathogens that cause gastrointestinal diseases worldwide. Apart from food contamination, fecal pollution has been consistently associated with the transmission of these pathogens, and their rapid detection in food and stools is of significance for food safety. Howev...
متن کاملDevelopment of a real-time PCR melt curve assay for simultaneous detection of virulent and antibiotic resistant Salmonella.
Multiple drug resistance in Salmonella is an emerging problem in the area of food safety. Depending on the virulence and antibiotic resistance characteristics of the Salmonella strain, infections of varying severity could result. In this study, a multiplex melt curve real-time PCR assay for the detection of virulent and antibiotic resistance strains of Salmonella was developed with two primer s...
متن کاملDetection of invasive fungal pathogens by real-time PCR and high-resolution melting analysis.
BACKGROUND/AIM Causative agents most frequently encountered in systemic infections are bacteria, although fungi that cause invasive infections have also emerged, mostly in immune-compromised patients. The early detection and adequate treatment of bloodstream infections are critical for successful treatment. The aim of this study was to develop a rapid and efficient method for the detection and ...
متن کاملOptimization and validation of a triplex real-time PCR assay for thermotolerant Campylobacter species associated with foodborne disease
The genus Campylobacter is globally recognised as the leading bacterial cause of human foodborne gastroenteritis. Every year around 8000 Swedes are infected by Campylobacter. Most people are infected by thermotolerant Campylobacter species, commonly C. jejuni and C. coli. In this study a triplex real-time PCR has been developed in order to be a last step in a qualitative method for identificati...
